Opinion is always protected as free speech. However text on this sign clearly accuses a specific, named individual of animal cruelty, a citable offense. It's a statement of fact which may or may not be true. If not true, it is defamation of character, and not protected as free speech.
If able to be proved to be true, one would expect the guilty party would be charged.
